Introduction: Nitric oxide NO ; is known to increase sodium excretion by inhibiting ENaC activities in the kidney. Previously we demonstrated that prostasin stimulates ENaC activities when the two are co-expressed in Xenopus oocytes. Therefore, we investigated the effect of NO on the prostasin in the regulation of ENaC activities in renal epithelial cells. Methods: We treated a mouse cortical collecting duct cell line M-1 cells ; with 500 M of NOC18 NO donor ; , 500 M of 8-bromo-cGMP, or 1 M of CAPE NF-KB inhibitor ; for 24 hours. mRNA and protein expression of prostasin were determined by real-time PCR and immunoblottings. We also examined the effect of CAPE or NOC18 on nuclear translocation of NF-KB in M-1 cells. Results: Treatment of M-1 cells with NOC18 significantly decreased mRNA and protein expression of prostasin by approximately 71 2.8 % and 68.6 2.1 % respectively. However, 8-bromo-cGMP had no effect on the expression of prostasin. The prostasin expression was also decreased by CAPE. NOC18, as well as CAPE, inhibited the nuclear translocation of NFKB in M-1 cells. Conclusion: These findings suggest the possibility that the NO-induced suppression of prostasin may play a role in the NO-mediated natriuresis in the kidney. The inhibition of prostasin expression by NO was not mediated by the classical cGMP pathways, but by the inhibition of the nuclear translocation of NF-KB in the renal epithelial cells.

However, this thesis rests on the belief that society is neither composed of all pre-programmed individuals or of all rational individuals. As long as some portion of the population is recognized as rational, analyzing cooperation in the finitely repeated Prisoners' Dilemma is necessary.
