Criteria for confluency were the epithelial resistance 300 Q cm 1 ; and accumulation of lactatc in the basolalcral compartment. Osmolytes were either measured with HPLC on a Sugar Pak I column or by using radioactive tracers. During hypotonic shock on both sides for 10 minutes a polarity of the organic osmolyte efflux pathways could be observed. Sorbitol, myo-inositol and'betaine were exclusively released basolatcrally 35, 15 and 48 % of intracellular content ; whereas glycerophosphorylcholine GPC ; and taunnc left the cell layer on both sides 48.l0.8% and 20.23.0% of total efflux apically ; . An apical release of taurine and GPC was also found in cell cultures grown in a one compartment culture system on gas-permeable membranes Petripcrm ; . Furthermore the different pathways of taurine release could be distinguished pharmacologically by their different sensitivity to so-called anion channel blockcrs such as D1DS, SITS and NPPB. We suggest that the apical pathway is a volume-sensitive anion channel of high SITS sensitivity used by taurine, chloride and perhaps other anions. The basolatcral system with a lower SITS and NPPB sensitivity is probably shared by taurine and myo-inositol because a diminished myo-inosilol efflux could be observed when both osmolytes were present intraccllularly at high amounts after preloading of the cells. These data indicate that volume regulation in IMCD cells during hypotonic shock is mediated via differently located but functionally coordinated efflux pathways.
