Bycatch precludes other more productive uses of fishery resources; it is particularly important to minimize the waste associated with bycatch when so many of the world's fisheries are either fully exploited or overexploited. Although not all discarded fish die, when bycatch becomes a source of fishing mortality it can slow the rebuilding of overfished stocks. Bycatch imposes direct and indirect costs on fishing operations by increasing sorting time and decreasing the amount of gear available to catch target species. In contrast, OTR concentrations in rabbit amnion cells are upregulated by agents that elevate intracellular cAMP concentrations and by glucocorticoids 7 ; . The concentrations of OTR in the mammary gland are relatively low at the end of parturition, when uterine and amnion receptor concentrations are greatest, but increase to maximal levels during lactation in the rat 12 ; . Because the temporal pattern of OTR expression differs between the mammary gland and uterus or amnion, it would seem that the mammary receptors are regulated differently than uterine amnion receptors. We recently demonstrated that a human carcinosarcoma cell line Hs578T ; expresses OTRs that are up-regulated by dextrancoated charcoal-treated FBS DCC-FBS ; 13 ; . Up-regulation was inhibited by pretreatment with the protein kinase C PKC ; inhibitor, GF 109203X 13 ; , indicating the possible involvement of AP-1 proteins. Because OTRs are not universally expressed and are regulated according to reproductive stage, it would seem that the OTR gene has important regulatory and or enhancer elements. To better understand OTR regulation in breast cells, we cloned the human OTR gene and expressed constructs containing the promoter region fused to luciferase in Hs578T cells. It has been demonstrated that calcitonin-binding sites are present in a variety of tissue types, including in the pituitary gland. Interleukin-6 IL-6 ; is also produced in the pituitary and it regulates the secretion of various hormones. In this study, we examined the expression of the calcitonin receptor and the mechanism of IL-6 production induced by calcitonin in the pituitary folliculo-stellate cell line TtT GF ; . The mRNA of calcitonin receptor subtype C1a, but not that of C1b, was detected by RT-PCR in TtT GF cells and in the normal mouse pituitary. Calcitonin increased cAMP accumulation and IL-6 production in a concentration-dependent manner in TtT GF cells. As calcitonin activates the PKA and PKC pathways, we investigated the contributions of PKA and PKC to IL-6 production. IL-6 production was only slightly increased by either 8-bromo-cAMP 1 mM ; or phorbol 12-myristate 13-acetate 100 nM ; alone. However, IL-6 was synergistically induced in the presence of both 8-bromo-cAMP 1 mM ; and phorbol 12myristate 13-acetate 100 nM ; . Furthermore, calcitonininduced IL-6 production was completely suppressed by H-89 PKA inhibitor ; or GF109203X PKC inhibitor ; , indicating that the activation of both PKA and PKC is necessary for calcitonin-induced IL-6 production. On the other hand, pertussis toxin Gi Go signaling inhibitor ; treatment achieved an approximately 9-fold increase in calcitonin-induced IL-6 production. These results show that calcitonin-stimulated IL-6 production is mediated via both PKA- and PKC-signaling pathways, whereas calcitonin also suppresses IL-6 production by activating Gi Go proteins in folliculo-stellate cells. BACKGROUND: Studies have suggested that ultrasound-guided embryo transfer UG-ET ; may improve the outcome in IVF; however, several factors may account for the improvement in pregnancy rate. This study examines the use of ultrasound to determine the accuracy of trial transfer TT ; in preparation for ET. METHODS: Sixty-seven consecutive patients prospectively underwent UG-ET over a 2 month period. Total cavity length by US was compared with the length noted by TT. A difference of cm was considered significant. All embryos were placed within 1 2 cm the fundus by US. RESULTS: Twenty patients 29.9% ; had a difference of cm and 13 patients 19.4% ; had a difference of # 1.5 cm. Patients with a difference of or .5 cm had a significantly greater depth at transfer P 0.001 ; and uterine cavity length P 0.001 ; when compared with patients without a difference. Clinical pregnancy, implantation, delivery and overall miscarriage rates did not differ between patients with a difference of or .5 cm versus no difference. There were no ectopic pregnancies. CONCLUSIONS: Nineteen percent of patients had a discrepancy of .5 cm and , 30% had a difference of cm from TT at UG-ET, suggesting a benefit to UG-ET. A large prospective randomized trial comparing UG-ET with blind transfer is required to assess further if UG-ET should be used in all cases of ET and hms.

Previously, we have found that stiffness increases with increasing applied stresses.1113 We wondered whether this phenomenon depended on the process of oxidative metabolism. Treating endothelial cells with potassium cyanide 2.5 mmol L for 15 minutes ; , an oxidative metabolism inhibitor, had no effect on inhibition of stress-induced stiffening response Figure 2 ; . A recent article suggests that stress-induced stiffening response might be dependent on tyrosine dephosphorylation.17 To test this possibility, we added RGD-coated beads for 15 minutes, washed away unbound beads, and then treated cells with a low dose of phenylarsine oxide PAO, 5 nmol L for 10 minutes ; . Surprisingly, inhibition of tyrosine phosphatase with PAO had no effect on stress-induced stiffening response Figure 3 ; . Treating the cells with a much higher dose of PAO 10 mol L for 10 minutes ; did not have an effect on the stiffening response either not shown ; . The reason for this discrepancy might lie in earlier experiments: the bead was added for seconds only before the force was applied.17 The focal adhesion complex was still in the process of being formed, ie, different linking proteins were still in the process of being recruited to the bead; thus, the forcedependent stiffening response observed in their experiments might reflect the strengthening process in the focal and hydromorphone.
