Other genes most likely involved in intracellular signaling cascades Stbx4, Wdr12 ; were also up-regulated by HA12. The majority of up-regulated genes were seen during active morphogenesis of HA12-stimulated endothelial cells 12 h; Table II and supplemental Table II ; . Among them, a marked and distinct up-regulation more than 3-fold induction ; was observed of genes encoding the transcription factors Nfyb and Pknox1; Nfyb and Pknox1 stimulate the transcription of various genes such as type 1 collagen 26 ; and urokinase 27 ; , respectively. Thus, the proteins induced upon HA12 stimulation of endothelial cells have broad roles in cell growth and homeostasis as well as in modulation of extracellular matrix and reorganization of the cytoskeleton. Is a rare disorder in which G aucher's disease-glucosidase is geneticallythe activity of lysosomal deficient, resulting in the accumulation of glucocerebroside in reticuloendothelial cells. The typical manifestations include hepatosplenomegaly and bone marrow infiltration with dysfunctional monocytes. Pulmonary hypertension has been described, although the etiology is unclear. We describe a patient with Gaucher's disease and pulmonary hypertension who has been successfully treated with longterm continuous IV epoprostenol. This is the first report of the successful treatment of pulmonary hypertension associated with Gaucher's disease, and it may have important implications for other patients with this condition.

Other studies have employed probes which are more specific for hyaluronan, such as the hyaluronan-binding region of the cartilage proteoglycan aggrecan or cartilage link protein, neither of which bind chondroitin sulfate. In an in vivo study of rat brain, hyaluronan was localized intraaxonally and within a select few nuclei in cerebellar neurons, as well as in the cytoplasm of granule cells10. The selectivity of the distribution of hyaluronan inside only a few cells made it unlikely that this was an artifactual finding. In an ultrastructural study of vascular tissue from rats, both link protein-gold and aggrecan-gold were used to morphologically localize hyaluronan in vivo. In endothelial cells and smooth muscle cells, hyaluronan was detected more prominently in caveolated portions of the membrane than on the naked cell surface11 Fig. 2 ; . These authors also detected hyaluronan in nuclear heterochromatin. The specificity of the staining in these studies was confirmed by abolishment of the labeling by digestion with Steptomyces hyaluronidase, which is highly specific for hyaluronan and hydroxychloroquine.

Fluorescein-hyaluronan fl-HA ; was prepared following a modification of the method described by de Belder and Wik 1975 ; . Briefly, 50 mg of hyaluronan HA ; Sigma, grade I ; was dissolved in 40 ml water, and mixed with 20 ml of DMSO. Fluoresceinamine, isotype I 25 mg, Aldrich ; in DMSO 0.5 ml ; containing acetaldehyde 25 l, Fluka ; and cyclohexyl isocyanide 25 l, Fluka ; was added into the mixture. The pH value of the solution was adjusted to 4.5. The mixture was incubated at 22C for 5 h, and then poured into 800 ml of 95% ethanol. The precipitated flHA was collected by centrifugation at 1, 000 g and air-dried in a chemical hood. The fl-HA was then dissolved in water and re-precipitated by 95% ethanol 2 times. The fl-HA was further purified by exhaustive dialysis against distilled water followed by lyophilization. The fl-HA was then characterized by Sepharose CL-2B chromatography and a proteoglycan-dependent ELISA. 3H-labeled hyaluronan [3H]HA ; was purified from the conditioned medium of rat fibrosarcoma cells labeled with [3H]acetate New England Nuclear ; as described previously Knudson and Toole, 1987; Nemec et al., 1987 ; . Following purification, the [3H]HA has been shown to be free of contaminating glycosaminoglycans as determined by DEAE chromatography Knudson and Toole, 1987; Nemec et al., 1987 ; and has a specific activity of 6.8104 cpm g and hydroxyurea.

D-Penicillamine D-Penicillamine is a cysteine-like amino acid slightly different chemical structure ; that has had several therapeutic uses. In Wilson's disease, D-penicillamine, which is also called "Cuprimine" Merck ; , has been given as an oral chelating agent for lowering blood copper levels.38 It has been found beneficial in some cases of rheumatoid arthritis where other modalities are ineffective.39 In cystinuria, D-penicillamine participates in sulfhydryl-disulfide exchange with cystine.40 This serves to reduce cystine levels by forming a cysteine-penicillamine disulfide which is excreted more readily in urine than cysteine. In heavy metal poisoning, D-penicillamine has been used for both lead and mercury removal, 41 , and it is also credited with removing arsenic.42 While removal of mercury, lead, copper and arsenic may sound like beneficial interventions for autistics, D-penicillamine also has possible, significant detrimental effects. First, it lowers cysteine levels by lowering blood cystine, and cysteine is often low or deficient to begin with in autism. By this process, D-penicillamine can also lower glutathione stores. Second, this drug is a pyridoxine antagonist. It complexes with the vitamin and impairs coenzyme function of vitamin B6 as pyridoxal 5-phosphate ; .43 And nearly 50% of autistics need and benefit from vitamin B6 with magnesium ; as a supplement.44 Pathological side effects from use of D-penicillamine include: sensitivities rash, fever ; , leukopenia, lymphadenopathy, proteinuria and nephritic syndrome, and a form of glomerulonephritis and immune dysregulation called Goodpasture's syndrome.8 While the severe side effects are uncommon, the sensitivity problem is common about one-third of users ; , and the cysteine and pyridoxine depletion are always occurring to some extent. Note also that all commercial forms of D-penicillamine are formulated to include lactose, 42 for which there is little tolerance in at least 60% of autistics. Therefore, D-penicillamine is not considered to be a preferred agent by DAN clinicians or researchers and hyaluronan.
