However, the regulation of the key transcription factor Egr-1 gene, by atherogenic factors in atherosclerotic lesions is little understood. Specifically, how the Egr-1 gene transcription is regulated in vascular smooth muscle cells SMC ; is currently unknown. Lysophosphatidic acid LPA ; is a component of oxidized lipoprotein and an agent released by activated platelets. Our data show that LPA markedly induces Egr-1 messenger RNA mRNA ; and Egr-1 protein in SMC. Results of the nuclear run-on experiments demonstrate that LPA-induced Egr-1 gene expression is principally controlled at the transcriptional level. Transfection studies using a series of deleted Egr-1 promoters revealed that a -140- to 21- base pair region of the Egr-1 promoter contains regulatory elements required for LPA-mediated induction. Electrophoretic mobility shift assays revealed that the DNA-binding activities of both cAMP-response element binding protein CREB ; and serum response factor SRF ; to the CRE and SRE motifs are markedly elevated in response to LPA, suggesting a role of both CREB and SRF in regulating LPA-induced Egr-1 expression. Transfection of site-directed mutants of the Egr-1 promoter demonstrated that CRE and SRE cis-acting elements in the Egr-1 promoter were required for LPA-induced Egr-1 gene induction. Our data show for the first time that lysophosphatidic acid induces Egr-1 gene expression in smooth muscle cells and that this Egr-1 gene expression is mediated by CREB and SRF transcription factors. Understanding the mechanisms of induction of the key transcription factor Egr-1 expression in response to atherogenic factors in vascular smooth muscle cells is important toward understanding the formation and development of atherosclerotic lesions and sulfasalazine.
